A linker containing stop codons in all three reading frames together with a floxed neomycin resistance cassette were introduced into exon 3. Targeting took place in ES cells homozygous for the Tg(Prm-cre)70Og transgene. Cre-mediated excision subsequently removed the neomycin cassette upon transmission from male chimeric mice. mRNA transcripts were produced but Western blot analysis demonstrated the absence of protein.
