A human GLI3 cDNA and a downstream loxP-flanked neomycin selection cassette were inserted in the first coding exon of the endogenous Gli2 locus. The inserted cDNA was flanked with an upstream FLAG epitope and a downstream mouse Gli2 3' UTR and three tandem repeats of a SV40 polyA signal. The level of GLI3 expression from this allele was reported to be less than that of the derivative allele, Gli2tm5.1(GLI3)Alj, from which the floxed neo cassette was removed. (J:87948)
