A cDNA encoding the entire RUNX1 protein was inserted into exon 4 of the endogenous locus via homologous recombination. Semiquantitative RT-PCR analysis of splenic and thymic message showed the levels of mutant transcript in homozygous mutant mice to be similar to the levels of normal transcript in wild-type mice.
