Exon 8 was replaced with a gene trap cassette inserted by homologous recombination. The targeting vector included a splice acceptor site, stop codons in all reading frames, beta-geo initiated by its own start codon, and a polyadenylation signal. Mutant transcript consisting of exons 1 through 7 and the beta-geo sequence was detected by RT-PCR analysis. Western blot analysis identified a truncated protein in homozyous mutant mice, putatively containing the MEIS-homology domain. Beta-geo expression was realized by staining. (J:87890)
