A floxed neomycin resistance gene was inserted in intron 3 and an additional loxP site was inserted in intron 4. Transient transfection of properly targeted ES cells with a Cre expressing plasmid resulted in excision of the neomycin gene leaving exon 4 flanked by two loxP sites. (J:86988)
