A genomic fragment encompassing exons 2, 3, 4, and 5 was replaced with a neomycin selection cassette inserted by homologous recombination. The deleted region encoded portions of the 5-methylcytosine binding and catalytic domains. Transcript was undetected by RT-PCR analysis of homozygous MEFs. (J:86981)
