A hypomorphic allele was generated by inserting an frt flanked neomycin selection cassette in the intron following exon B. The targeting vector also included a proline to alaline missense mutation at codon 12 (P12A) and loxP sites flanking exon B and the neo cassette. mRNA corresponding to both alternatively spliced transcripts 1 and 2 were significantly reduced in homozygous mutant white adipose tissue. While variant 2 transcript was nearly undetected in homozygous mutant brown adipose tissue, Variant 1 was significantly increased relative to wild-type. (J:86705)
