The ACT codon for threonine at amino acid residue 72 was changed to a GCT alanine codon. At the same time, the CCC codon for amino acid residue 73 was changed to CCA which eliminated a Pst 1 restriction site but did not have any other effect. Finally, a floxed neomycin gene was inserted in intron 4. The neomycin resistance gene was removed in vivo by mating to mice expressing Cre recombinase from the protamine promoter in the male germ line.
