A 2.1 kb genomic fragment encompassing the exons encoding the fourth transmembrane domain was replaced with a neomycin selection cassette inserted by homologous recombination. Neither transcript nor protein was detected by RT-PCR or Western blot analyses of homozygous mutant brain stem tissue. The absence of protein was also verified in fractions obtained from mutant spinal cord extracts. (J:86625)
