The Rag1 coding sequence was replaced with a truncated core Rag-1 sequence followed by a floxed neomycin resistance gene. The neomycin gene was then removed from properly targeted ES cells by transient transfection with a plasmid based Cre recombinase. Core Rag1 was expressed at levels comparable to that found for intact protein. (J:86464)
