The exon encoding the biotin binding motif and flanking genomic sequence was replaced by an HPRT expression cassette by homologous recombination. Northern blot analysis on RNA derived from skeletal muscle demonstrated that no detectable transcript was expressed from this allele in homozygous mice. Western blot analysis on extracts of various tissues confirmed that no detectable protein was present in homozygous mice. (J:68427)
