This transgenic construct was generated by ligating sequence encoding a fusion protein of cre recombinase and a mutant mouse estrogen receptor ligand binding domain to a CMV early enhancer and chicken beta-actin promoter. In the absence of ligand binding, the fusion protein remains in the cytoplasm. Binding of 4-hydroxytamoxifen enables the fusion protein to move to nucleus where it may act upon DNA. This transgenic line was generated by electroporating the contstruct into ES cells. Expression of the fusion protein was in a widespread mosaic pattern. Two lines were generated, BD8 with two copies of the transgene and CA6 with one copy. (J:81562)
