Floxed sequence encompassing exon 3 and and FRT-flanked neo cassette was excised from Ptchtm1Bjw via in vivo cre-mediated recombination. Truncated transcript lacking the deleted region was detected in mutant mice by RT-PCR analysis. The mutant transcript encodes the first 117 residues followed by 24 missense residues and a nonsense mutation, putatively resulting in a protein truncated ~13 residues after the first transmembrane domain.
