Exon 2 was replaced with a cassette containing EGFP followed by floxed neo. GFP was expressed by the endogenous promoter and regulatory elements, however, comparison with endogenous expression and with Serpinb6atm1.1Pib GFP expression indicated that retention of the neo cassette increased expression levels. Western blot analysis indicated an absence of normal protein in homozygous mutant spleen and thymus tissue. (J:85039)
