A floxed neomycin selection cassette and an additional loxP site were inserted at the endogenous locus by homologous recombination. The neomycin selection cassette was deleted in ES cells by transient expression of Cre recombinase, resulting in loxP sites flanking the promoter region, the transcription initiation site, and exon 1 in the final allele.
