Both metallothionein I and II were disrupted using the same targeting construct. The second exon of metallothionein II had an insertion of the pMC1NeoPA selection marker at a position 30bp downstream of the start codon. Northern blot analysis of liver mRNA from homozygotes in which the gene was induced by intraperitoneal injection of copper demonstrated that transcription was prevented. Further, no protein product was detected in the livers of these animals. (J:14416)
