Exon 8 was replaced with a cassette containing a splice acceptor site, an IRES, and sequence encoding a bifunctional lacZ-neomycin fusion protein. The insertion was designed to disrupt the expression of both isoforms that result from alternative splicing of the 5' region. Western blot analysis of the frontal cortex and striatum indicated an absence of normal protein in homozygous mutant mice. (J:84278)
