A cassette expressing the neomycin resistance gene and the thymidine kinase gene flanked by loxP sites was inserted into intron 2. An additional loxP site was placed in intron 3. Homologous recombination was confirmed by Southern blots. The insertion of a neoTK cassette into intron 2 of the Men1 locus (the T allele) disrupted the Men1 expression at RNA and protein levels, as shown by RT-PCR and Western blot, respectively. (J:83882)
