The coding sequence of exon 1 was replaced with an EGFP-neo cassette which caused the termination of transcription. The neomycin resistance gene was flanked by loxP sites. Correct targeting was demonstrated by Southern blot. Transcripts were lacking in northern blots of total liver RNA from mice homozygous for the targeted allele and no Hsf2 protein expression was seen in western blots. (J:83730)
