Flanking loxP sites and a PGK-neo cassette were introduced to the exons encoding the transmembrane domains. These exons were subsequently deleted by transient expression of cre recombinase in correctly targeted ES cells. Absence of gene expression in homozygous mutant embryos was confirmed by Western blot analysis. (J:83476)
