The wild-type mouse Hbegf cDNA, including the polyadenylation signal and flanked by loxP sites, is joined to the beginning of the first exon of the endogenous gene so that its expression is driven by the native promoter. The rest of exon 1 through exon 3 and most of intron 3 have been deleted. Downstream of the Hbegf cDNA are the bacterial beta-galactosidase gene (lacZ), with a polyadenylation signal and a nuclear targeting sequence, and the neomycin resistance gene driven by the phosphoglycerate kinase promoter, also with polyadenylation signal (Pgk-neo cassette), separated by a short segment from the middle of the third intron of the native gene.
