The endogenous EDA exon, which is subject to alternative splicing, was replaced with a floxed EDA exon modified with point mutations for optimization of both the 5' and the 3' splice sites. RNase protection and RT-PCR analysis confirmed constitutive EDA exon inclusion, showing the EDA exon to be included in 99-100% of transcript in all tissues of homozygous mutant mice. (J:84274)
