Gene targeting was used to flank exon 1 with a floxed PGK-neo cassette on one side and a loxP site on the other side. Correctly targeted ES cells were transiently transfected with a cre-recombinase plasmid to delete exon 1. Absence of gene expression in homozygous mutant animals was confirmed by Northern blot and RT-PCR analysis. (J:82471)
