The 5' two-thirds of the endogenous coding region was replaced with a floxed neo cassette inserted by homologous recombination. Normal transcript was undetected by Northern blot and RT-PCR analyses of both homozygous mutant mice and heterozygous mice carrying a paternal mutant allele. An absence of protein was verified in these mice by Western blot analysis of brain tissue. (J:66557)
