The gene was disrupted by replacement of exon 1 with a diphtheria toxin A (DTA) chain gene and a floxed neomycin resistance cassette. The exon 1 splice site is situated 3' to the neo cassette. The floxed neo cassette was removed by transient expression of cre recombinase in correctly targeted ES cells, thus allowing transcription from the DTA gene. (J:71355)
