Flanking loxP sites were introduced to the large exon and a floxed neo cassette was inserted into the upstream intron. Transient expression of cre recombinase in correctly targeted ES cells resulted in deletion of the floxed neo cassette leaving behind loxP sites flanking the large exon. Mutant animals were then mated to cre deleter strain Tg(ACTB-cre)13Miya to generate null animals. (J:81816)
