The endogenous locus was replaced with a floxed neo cassette followed by cDNA sequence encoding a mutant truncated RAG2 protein ('core' RAG2) lacking the C-terminal 144 amino acids. The floxed neo cassette was subsequently removed by transient expression of cre recombinase in correctly targeted cells. (J:81837)
