Exon 2 of the endogenous locus was replaced with a cassette containing Beta-gal and neo genes, resulting in an isoform-specific mutation. Exon 2 is the second coding exon of the Citron-K isoform and is far upstream of the Citron-N trascriptional start site. Western blot analysis of cerebral, splenic, and thymic tissues inidicated an absence of the Citron-K isoform and showed normal levels of the Citron-N isoform in homozygous mutant mice. (J:77297)
