A cDNA fragment encoding the runt domain and N-terminus of mouse Runx2 (beginning at exon 2) was cloned between the promoter and enhancer elements of Col2a1 to produce a dominant negative transgene. The Col2a1 promoter is active in chondrocytes. Transgene expression was confirmed by Northern blot analysis of RNA extracted from the head or trunk of day E12.5 transgenic embryos. (J:68793)
