The Lck promoter was observed to drive expression in the T-cell containing tissues including the thymus, spleen and lymph nodes. The highest levels of expression were observed in thymic tissue. Immunohistochemical analysis of T-cell containing tissues confirmed overexpression of a dominant negative peptide consisting of the DNA binding domain (residues 1 through 376) of the human ADP-ribosyltransferase protein. ADP-ribose polymerization was undetected in thymocytes treated with MNNG, a monofunctional alkylating agent, indicating inhibition of the enzymatic activity of the endogenous protein. (J:80883)
