Sequence encompassing exons 2 through 5, encoding amino acids 6 through 274, was replaced when a PGK-hprt cassette was inserted at the endogenous locus via homologous recombination. The deleted region included all but 5 residues of the prodomain and 8 residues of the catalytic domain. Northern blot analysis using probes for the amino- and carboxyl-terminal untargeted regions indicated a complete absence of transcript produced from the targeted allele. (J:57969)
