A neomycin selection cassette was inserted into exon 3. The insertion disrupted the coding region 12 codons downstream of the erythroid-specific start codon. While no protein was detected in red blood cells obtained from homozygous mutant mice, translation of the kidney specific isoform, which is initiated from a start codon downstream of the inserted neo cassette, was verified by Western blot analysis of kidney lysates.
