The transgenic construct contained murine Cdkn1a cDNA sequence adjoined to murine transthyretin (Ttr) sequence including 3 kb of the upstream regulatory region, exon 1, intron 1, and a portion of exon 2. Both the endogenous start codon and an additional ATG in exon 2 of Ttr were converted to restriction sites by site-directed mutagenesis. The Ttr regulatory region directs expression in the liver.
