A sequence fragment containing portions of exon 7 and 8 was replaced with a neo cassette inserted by homologous recombination. The deleted region encoded the majority of the SH2-like domain. Flow cytometry and the use of labeled thymidine indicated that the mutant protein localized to the surface of thymocytes but did not transduce signals. (J:31167)
