A tissue specific transgene encoding an inducible MYC protein was generated by coupling the human involucrin promoter to a fusion cDNA encoding the human MYC protein joined to a modified mouse estrogen receptor (ER) ligand binding domain. The Gly525Arg mutant ER domain ("ERTM") responds to 4-hydroxytamoxifen (4OHT) but is insensitive to the endogenous ligand, 17beta-estradiol; therefore, MYC is active only in the presence of 4OHT. Immunohistochemical analysis with antibodies vs. either MYC or ER detected expression of the fusion protein in suprabasal keratinocytes within the intrafollicular epidermis and in the inner root sheath of hair follicles.
