The second coding exon of the endogenous gene was disrupted by homologous recombination of a construct containing IRES-lacZ and a floxed neo gene and subsequent excision of the selection cassette. A single loxP site was left downstream of the lacZ reporter, which was expressed via the endogenous promoter. RT-PCR analysis of homozygous mutant mice showed a lack of normal transcript. (J:79367)
