Embryonic stem cells carrying a deletion for the HsA enhancer element (Rr282726) were subjected to gene targeting to replace the Ecgamma 1 enhancer element (Rr275254) with a loxP site flanked neomycin resistance gene cassette. Correctly targeted cells, in which the Ecgamma 1 enhancer element was mutated on the same chromosome as the HsA deletion, were transfected with a cre expression plasmid to remove the floxed neo cassette.
