The endogenous locus was disrupted by the insertion of a vector that replaced the 3' and 5' splice sites of exons 1 and 2, respectively, with sequence containing stop codons. The targeting vector also included a neo cassette that replaced the majority of intron 1. Neither transcript nor protein was detected by Northern or Western blot analyses of brain tissue obtained from homozygous mutant mice. (J:79368)
