The exon encoding the ATP binding region of the kinase domain was replaced by a neo cassette inserted by homologous recombination. The absence of normal trancsript and normal protein was confirmed by Northern and Western blot analyses. In vitro studies showed normal proliferation of fibroblasts derived from homozygous mutant mice after infection with a retrovirus containing the wild-type cDNA. (J:74580)
