An exon encoding 51 N-terminal amino acids comprising most of the RING finger domain was deleted and replaced with a PGK-neo cassette via homologous recombination. Western blot analysis of correctly targeted ES cells showed no detectable protein expression. Homozygous mutant embryos were observed at day E4.0 by PCR genotype analysis. (J:69926)
