Exons 2 and 3 were replaced by a cassette containing lacZ and neo inserted by homologous recombination. The deleted region encodes the microtubule binding and bundling properties of the protein. Western blot and immunohistochemical analyses showed an absence of protein in hemizygous mutant male mice and reduced levels in heterozygous mutant female mice, indicating that protein is not produced by the targeted loci. Expression of the lacZ gene was driven by the endogenous promoter. (J:78746)
