The gene was disrupted by replacement of part of the promoter region and sequences encoding the first 21 amino acids with a neomycin resistance cassette. Northern blot using a probe to the C-terminal region of the gene and RT-PCR analysis of homozygous animals confirmed inactivation of the gene. Sequence analysis of a weak RT-PCR product revealed an alternatively spliced truncated transcript. (J:71804)
