A cDNA fragment encoding the intracelluar portion of the human RET51 isoform was fused in-frame with mouse exon 11 immediately downstream of the transmembrane domain-coding region. A floxed PGK-neo cassette followed the human cDNA sequence for positive selection. RT-PCR analysis and Western blot using allele-specific primers and antibodies, respectively, confirmed expression of the chimeric gene product in homozygous mutant animals.
