Parts of introns 1 and 2, and regions of exon 2, which contains the translation initiation sites, were replaced by a neomycin resistance gene via homologous recombination. Absence of gene expression in adipose tissues of homozygous mutant animals was confirmed by Northern blot analysis. Protein product was undetectable in plasma of homozygous mutants. (J:77479)
