A construct containing human exon 5 followed by the entire human CBFA2T1 coding region was inserted at the endogenous locus via homologous recombination, replacing the endogenous exon 5. The targeted allele generates a fusion protein similar to the fusion protein resulting from T(8;21) which is associated with the M2 subtype of acute myeloid leukemia (AML). Transcription of the allele was verified in heterozygous mice by a protection assay. (J:38705)
