The gene was disrupted by replacing a 0.15 kb region, which includes part of exon 6 and the downstream intron, with a neomycin resistance cassette. The mutation was confirmed by PCR analysis using primers to the neo insertion. Absence of gene expression in homozygous mutants was demonstrated by Western blot analysis of splenocyte lysates.
