The entire coding region of the gene (from the translation initiation codon to 463 bp of the 3' flanking region) was replaced with a neomycin resistance cassette via homologous recombination. Absence of gene expression in homozygous mutant animals was demonstrated by RNase protection assay and Western blot analysis. Furthermore, in situ hybridization and immunohistochemical analysis of testes from homozygous mutant animals confirmed the absence of RNA and protein products. (J:60631)
