A cassette containing an IRES-lacZ gene followed by a floxed neo gene was inserted into the first coding exon of the endogenous locus via homologous recombination. Transcript was undetected in homozygous mutant mice by Northern blot analysis of limb cartilage and RT-PCR analysis of cartilage, thymus, and whole eye tissue. Immunohistochemical analysis of postnatal tibial growth plates confirmed the absence of protein. (J:78596)
