Exon 2 was replaced with a floxed PGK-neo cassette via homologous recombination. Prior to blastocyst injection, the PGK-neo cassette was removed by transient expression of cre recombinase in correctly targeted ES cells. Southern blotting confirmed correct targeting of construct. (J:64863)
