Seven copies of the tetracycline operator were inserted in the 5' untranslated region of the endogenous locus via homologous recombination. An RNAse protection assay revealed the inserted sequence to be approximately 100 bp upstream of the transcriptional start site. While quantitative analysis of the amount of transcript produced by the mutant allele showed a 55% reduction relative to the wild-type allele, in situ hybridization indicated that the pattern of expression was unaffected.
